Enzymes: Catalase

Week 1:  The study of ENZYME CONCENTRATION

Solutions and Other Consumables made ahead of time:
Potato Extract
(contains the enzyme of study, catalase).    Make just before the lab.  Combine 250g of potato with  100 mL of phosphate buffer in blender.  Filter through a layer of cheese cloth and wire strainer.  Each group will need 10mL aliquoted for their own use. (10mL/group x ~6 groups/lab)

0.1M phosphate buffer –Each group will need 10mL buffer aliquoted for their own use.

Make anytime and aliquot into 10mL tubes.  (10mL/ group x ~6 groups/lab)

1% Hydrogen peroxide  (80mL/group x ~6 groups/lab = 480mL)

Week 2:  The study of SUBSTRATE CONCENTRATION

Solutions and Other Consumables made ahead of time:
Potato Extract
(contains the enzyme of study, catalase).    Make just before the lab.  Combine 250g of potato with 100  mL of phosphate buffer in blender.  Filter through a layer of cheese cloth and wire strainer.  Each group will need 10mL aliquoted for their own use. (10mL/group x ~6 groups/lab)

Various H2O2 Concentrations:  Made from 3% hydrogen peroxide for a total volume of 500mL for each of the concentrations below-
0.3%,  0.6%,  0.8%,  1.0%,  1.5%, and  2.0%

6  glass vials
10mL pipette tip with ratchet dispenser
10 ml of 100% potato extract
10 mL of 0.1M phosphate buffer (
20-200 uL micropipettors and tips
200-1000 uL micropipettors and tips
8 Microcentrifuge tubes
Sponge to hold microcentrifuge tubes
Black marker
Stop watch
Ice/plastic beakers for ice
10mL of Potato extract –need holder for the tube
10mL of phosphate buffer- need holder for the tube

(100mM Phosphate Buffer ( 1 liter - 8.70 K2HPO4 dibasic; 6.80 KH2PO4 ))
